Liquisolid technique is an emerging method of formulation development. In the present work, modified release formulation of gliclazide was developed by using liquisolid technique. Gliclazide was dispersed in a nonvolatile solvents system of Tween 80 and Propylene glycol followed by adsorbing the liquid medication on carrier material MCC pH 102. Drug release rate controlling polymers, HPMC were wet granulated with binder solution along with diluent. The two blends of API liquidsolid and polymer mix, were blended uniformly with extra granular excipients to manufacture Gliclazide Modified Release tablets 30 mg. Characterization of gliclazide liquisolid tablets was done using analytical techniques i.e. FTIR and XRD. The formulation showed zero order release pattern and matched with the Innovator (Diamicron MR 30 mg) as well. The optimization of the formulation was done by using Mixture Design where amount of Propylene Glycol, Tween 80, DCP (anhydrous), HPMC K4M, HPMC K100 LVCR were considered to be the independent formulation variables and dissolution at respective time points, and hardness were considered as dependent variables. The formulations were evaluated for dissolution, assay, hardness, thickness and other important properties. Dissolution profile of proposed formulation was found comparable to Innovator tablets. The modified release tablets showed satisfactory stability at 40 °C / 75 % RH for a period of 3 months.
