COMBINED MIXTURE DESIGN IN OPTIMIZATION OF EXTRACTION SOLVENT IN ASSAY TEST OF ETHYL CELLULOSE COATED TABLETS

The aim of the present study was to apply design of experiments to optimize extracting solvent in assay method of ethyl cellulose coated tablets. For this purpose, tramadol HCl extended release tablets were selected with combination of aqueous and organic solvents as extraction solvent for assay method development. Combined mixture design was implemented to optimize extraction solvent composition to enhance filtration volume and recovery of assay test sample. Solvent composition, centrifugation time and additional filtration step were evaluated as variables. Filtration volume through whattman No:1 filter paper and 0.45 micron PVDF filter were selected as responses. Results indicated that solvent composition and inclusion of additional filtration step enhanced filtrate volume and assay recovery. Conclusively design of experiments as an effective tool for optimization of extraction solvent in assay method development was established and validated. The proposed method has adequate reproducibility and accurate for estimation of tramadol HCl in ethyl cellulose coated tablets.
