Simultaneous Determination of Paracetamol, Aceclofenac with Tramadol Hydrochloride in Pharmaceutical Dosage Form by RP-HPLC Method

Chromatographic separation of paracetamol, aceclofenac and tramadol hydrochloride were performed on a Chromatopak C-18 column (25 cm x 4.6 i.d. x 5µm) as stationary phase with a mobile phase composed of phosphate buffer pH 7.0: acetonitrile (65:35, v/v), pH 7.0 adjusted with 5℅ triethyl amine at a flow rate of 1ml/min, detection was carried out at 225 nm. The retention time of paracetamol, aceclofenac and Tramadol hydrochloride were found to be 2.7 min, 4.2 and 6.0 min, respectively. The proposed method was validated for linearity, accuracy, precision, LOD and LOQ. Linearity, accuracy and precision were found to be acceptable over the ranges of 10-60 µg/ml paracetamol, 1-30 µg/ml for aceclofenac, 1-7µg/ml for tramadol hydrochloride.
