APPLICATION OF LIQUISOLID TECHNIQUE FOR ENHANCEMENT OF DISSOLUTION OF A WATER INSOLUBLE DRUG -CHOLECALCIFEROL

Most of the oral drug delivery candidates possess poor solubility and bio availability. Enhancing the solubility and bioavailability of such drugs have become a major challenge. Liquisolid technique, a newly developed approach for dissolution enhancement can circumvent these barriers. The aim of the present work is to enhance the solubility of poorly soluble drug cholecalciferol by using liquisolid technique. The solubility studies of cholecalciferol were performed in mixture of Polysorbate 80 and PEG 400 in different ratios (1:1, 1:2, 2:1, 1:3 and 3:1). Maximum solubility was observed in 1:2 ratio. The optimization was carried out by using Plackett Burman Design (Design Expert software). Change in X-ray diffractogram and shifting of endothermic peak in DSC from 85 ⁰C to 146 ⁰C, indicated complete conversion of drug into amorphous form in liquisolid tablets. Drug release profile of In-house developed formulation is better than the generic product and is a comparable to Divisun tablets 2000 I.U (Innovator).
